This 7.3 kb vector harbors the cap and rep genes that make the capsid and DNA replication proteins required to make infectious virions. This vector is cotransfected into the packaging cell line for production of recombinant, replication-deficient AAV virions.The pAAV-RC plasmid contains the AAV-2 rep and cap genes, encoding replication proteins and viral capsid structural proteins, respectively. Establishing the proper expression levels of rep and cap gene products is an important step in achieving high-titer virus production. pAAV-RC directs Rep and Cap expression from two different promoters, achieving optimal expression levels for each set of gene products.
